  ►    Mechanism of Drug Drug Interaction :  Dofetilide should not be given with other drugs that prolong the QT interval. Class I or class III antiarrhythmics should be stopped at least 3 half-lives before dofetilide is given. Potassium-depleting diuretics may cause hypokalaemia or hypomagnesaemia, increasing the potential for torsade de pointes, Dofetilide is metabolised to a small extent by the cytochrome P450 isoenzyme CYP3A4, and drugs or foods that inhibit this isoenzyme, such as macrolide antibacterials, HIV-protease inhibitors, diltiazem, and grapefruit juice, should be used with caution. Cimetidine, trimethoprim, ketoconazole, prochlorperazine, and megestrol, should not be given as they inhibit the renal excretion of dofetilide; verapamil is also contra-indicated as it too may substantially increase dofetilide concentrations
