  ►  Mechanism of Action :   Heparin’s mechanism of action depends on the presence of a specific plasma protease inhibitor, antithrombin III. Antithrombin III is actually a misnormer because, in addition to inactivating thrombin, antithrombin III inactivates other serine proteases including factors IXa, Xa, XIa, and XIIa. Antithrombin III can be considered as a stoichiometric “suicide trap” for these serine proteases. When one of the proteases encounters an antithrombin III molecule, the serine residue at the active site of the protease attacks a specific Arg-Ser peptide bond in the reactive site of the antithrombin. The result of this nucleophilic attack is the formation of a covalent ester bond between the serine residue on the protease and the arginine residue on the antihtrombin III. This results in a stable 1 : 1 complex between the protease and antithrombin molecules, which prevents the protease from further participation in the coagulation cascade.,
