►    Mechanism of Drug Drug Interaction :  Probenecid prolongs the half-life of Procaine benzylpenicillin by competing with it for renal tubular secretion and may be used therapeutically for this purpose. Antagonism has been reported to occur with some bacteriostatic drugs, such as chloramphenicol or tetracyclines, that interfere with active bacterial growth necessary for procaine penicillin to achieve its effect. And both enhancement and antagonism of its activity have been demonstrated for beta-lactam combinations. ,

►  Contraindication : Hypersensitivity, renal impairment, penicillin-sensitive patient,

►  Mechanism of Action :   It inhibits the bacterial growth by interfearing with the transpeptidation reaction of bacterial cell wall synthesis. Procaine benzylpenicillin is a structural analog of the natural D-Ala-D-Ala substrate, covalently bind to the active site of PBPs. This inhibits the transpeptidation reaction, halting peptidoglycan synthesis, and the cell dies.,
